The endothelin system. A new target for therapeutic intervention.
نویسنده
چکیده
Since its identification in 1988,1 endothelin has attracted intensive interdisciplinary research interest because of its unique profile as an endothelium-derived vasoactive factor with a powerful and characteristically long-lasting vasopressor activity. Indeed, in terms of cellular mechanisms of action, endothelin appears to be quite similar to many classic vasoconstrictor substances such as angiotensin II and norepinephrine. However, the slow time course of the regulatory mechanisms of its biosynthesis and secretion more resembles that of inflammatory cytokines, giving this family of small peptides a unique position within the realm of intercellular mediators with cardiovascular relevance.

Apelin is a recently discovered bioactive peptide, known to be an endogenous high-affinity ligandfor the previously orphan G protein-coupled receptor APJ. Apelin/APJ as a novel signaling pathwayhas been shown to play many crucial roles in cardiovascular function, blood pressure regulation, fluidhomeostasis, feeding behavior, obesity, type 2 diabetes mellitus, adipoinsular axis regulation, cellp...
متن کاملEvidence for the endothelin system as an emerging therapeutic target for the treatment of chronic pain
Many people worldwide suffer from pain and a portion of these sufferers are diagnosed with a chronic pain condition. The management of chronic pain continues to be a challenge, and despite taking prescribed medication for pain, patients continue to have pain of moderate severity. Current pain therapies are often inadequate, with side effects that limit medication adherence. There is a need to i...
متن کاملInterplay of Phosphorylated Apoptosis Repressor with CARD, Casein Kinase-2 and Reactive Oxygen Species in Regulating Endothelin-1–Induced Cardiomyocyte Hypertrophy
Objective(s): The role of the Apoptosis repressor with caspase recruitment domain (ARC) in apoptosis and in certain hypertrophic responses has been previously investigated, but its regulation of Endothelin-1 induced cardiac hypertrophy remains unknown. The present study discusses the inhibitory role of ARC against endothelin–induced hypertrophy. Results:In present study Endothelin treated car...
متن کاملRoles for endothelin receptor B and BCL2A1 in spontaneous CNS metastasis of melanoma.
Metastatic spread of melanoma to the central nervous system (CNS) is a common and devastating manifestation of disease progression, which, despite its clinical importance, remains poorly understood with respect to underlying molecular mechanisms. Using a recently developed preclinical model of spontaneous melanoma CNS metastasis, we have identified alterations in expression of endothelin recept...
متن کاملNEP, ACE and Homologues: The Pathophysiology of Membrane Metalloproteases
The zinc metalloprotease, neprilysin (NEP), plays a role in the metabolism of cardiovascular, inflammatory and neuropeptides, including mitogenic peptides such as bombesin. In the cardiovascular system, NEP has a primary role in the inactivation of natriuretic peptides but also contributes to local metabolism of angiotensin, endothelins and bradykinin. Hence NEP is seen as a potential therapeut...
